Layers of statistical modelling: the case of insulin sensitivity.

Abstract.
As applied statisticians, we meet problems that contain elements of
Deterministic Modelling, Stochastic and statistical Modelling,
Parameter Estimation, Numerical Methods, Population modelling (mixed
models), Optimization and of course Statistical Inference. This will
be illustrated using Diabetes mellitus data. In patients having type 2
diabetes, the body fails to respond to insulin resulting in low
"insulin sensitivity". In this talk, measurements from Intra Venous
Glucose Tolerance Test (IVGTT) for both healthy subjects and type 2
diabetes patients are used together with Bergman's deterministic
minimal compartment model (ODE) to estimate the insulin sensitivity
based on a nonlinear mixed effects model. Different sources of
variation are explored and an attempt is made to extend the
deterministic minimal model to a stochastic differential equation
(SDE) model to improve the performance and to get better parameter
estimates. Similar ideas can be used in other situations involving
compartment models such as Pharmacokinetics experiments.
